Histology & Histopathology Methods for TNBS Induced Colitis in Mice

TNBS Colitis Scoring

Multiple sections of H&E and PAS stained colon on slides, all sections present used for analysis using criteria below and scored for inflammation, necrosis and PAS staining.

Inflammation

Neutrophil and macrophage infiltration predominantly, fibroplasia and neovascularization in areas of transmural necrosis.

0 = No inflammation
0.5 = Very minimal focal infiltrates in mucosa only, affects <2% of the total colon
1 = Minimal focal or multifocal infiltrates in mucosa only, affects 2-10% of the total colon in area of necrosis or is minimal and diffuse throughout the mucosa if there is no necrosis
2 = Mild focal or multifocal infiltrates affecting mucosa, submucosa, outer muscle layers and serosa, affects 11-25% of the total colon in area of necrosis and is minimal to mild throughout the rest of the mucosa or mild diffuse throughout the mucosa if there is no necrosis
3 = Moderate focal or multifocal infiltrates affecting mucosa, submucosa, outer muscle layers and serosa, affects 26-50% of the total colon in area of necrosis and is mild throughout the rest of the mucosa or moderate diffuse throughout the mucosa if there is no necrosis
4 = Marked multifocal to diffuse infiltrates affecting mucosa, submucosa, outer muscle layers and serosa, affects 51-75% of the total colon in area of necrosis and is mild to moderate throughout the rest of the mucosa
5 = Severe multifocal to diffuse infiltrates affecting mucosa, submucosa, outer muscle layers and serosa, affects >75% of the total colon in area of necrosis and is moderate to marked throughout the rest of the mucosa

Necrosis

Measure the total length of all colon sections, measure the total length of necrotic mucosa devoid of epithelium to determine % area affected.

0 = No necrosis
0.5 = Very minimal, focal, affects <2% of the total colon
1 = Minimal focal or multifocal, affects 2-10% of the total colon
2 = Mild, focal or multifocal, affects 11-25% of the total colon
3 = Moderate focal or multifocal, affects 26-50% of the total colon
4 = Marked focal or multifocal, affects 51-75% of the total colon
5 = Severe, affects >75% of the total colon

PAS Staining for Mucous in Colonic Goblet Cells

0 = Normal glands with normal diffuse PAS staining
0.5 = Very Minimal decrease, affects <2% of the total colon
1 = Minimal focal or multifocal decrease, affects 2-10% of the total colon
2 = Mild focal or multifocal decrease, affects 11-25% of the total colon
3 = Moderate focal or multifocal decrease, affects 26-50% of the total colon
4 = Marked focal or multifocal decrease, affects 51-75% of the total colon
5 = Severe decrease, affects >75% of the total colon
